摘要
目的 :进一步明确葛根素改善微循环作用是否与封闭白细胞表面粘附分子LFA 1(CD11a/CD18)结合位点有关。方法 :12 0例急性脑梗塞患者于葛根素治疗前后取外周血分离多形核粒细胞 (PMN) ,应用间接免疫荧光标记 ,流式细胞仪检测白细胞表面粘附分子CD11a、CD18表达阳性百分率 ;应用透射电镜观察外周血PNM超微结构变化 ,并与 5例正常对照组比较。结果 :葛根素能明显降低外周血白细胞CD11a、CD18免疫阳性细胞数 ,抑制PMN代谢。结论 :葛根素具有封闭白细胞表面粘附分子CD11a、CD18结合位点 ,阻断白细胞与血管内皮细胞粘附 ,抑制PMNs活性 ,保护神经细胞的作用。
Objective: To investigate whether the microcirculation of Puerarin was related to its inhibition of WBC adhesion to endothelium of microvessels.Method: 120 adult patients were randomly divided into Puerarin group and control group.The percentage of CD11a and CD18 expression originated from polymorphonuclears(PMNs) were measured with indirect immunofluorenscence and flow cytometry,PMNs ultrastructure of peripheral blood were observed by the transmission electron microscope.Results: Puerarin coud decrease the numbers of immunopositive CD11a and CD18 cells in blood.PMNs ultrastructure on electron micrcoscopes were showed that PMN metabolism of Puerarin group less active compared with control group.Conclusion: The microcirculation improvement of Puerarin was related to is inhibition of WBC adhesion to endothelium of microvessels.
