肝脏纤维化发生机制的研究

摘要 肝脏纤维化的发生机制是近年来肝脏疾病研究的重点之一。目前的研究证明肝肛的星形细胞是肝纤维化时肝内胶原和其它细回外基质的主要来源。在肝纤维化过程中星形细凹逐渐转化为肌纤维母细胞而产生大量的细胞外基质沉积于肝脏。肌纤维母细胞的收缩可能与门脉高压的形成有一定关系。星形细胞的转化受TGF β等许多细胞因子及其它因素的激活。星形细胞合成胶原受多种水平的调节,转录后的调节受α-CsP与3’UTR的结合和蛋白因子与5’干环结构结合两方面的影响,合成后的饲节受基质金属蛋白酶及其组织抑制物的调节。另外,增生的胶原对星形细胞的反馈抑制功能丧失及肝窦的毛细血管化也对肝纤维化的形成有促进作用。
