摘要
目的自从建立起甲、乙、丙、丁、戊5种肝炎病毒的病原学诊断之后,仍有少部分肝炎患者的病因得不到明确,因此不少学者试图探索是否还有新型肝炎病毒的存在,并进行了大量的流行病学和实验诊断的研究,认为的确存在可经肠道外传播并引起人类肝炎的致病因子,目前关于庚型肝炎病毒(HGV/GBV-C)的致病性和组织嗜性尚无结论性资料我们研究了HGV/GBV-C与HCV混合感染者PBMC和肝脏中HGV/GBV-C复制中间体(负链RNA)的存在状况。方法应用逆转录巢式PCR技术,检测了32例肝炎患者PBMC和肝脏中HGV/GBV-C和HCV正负链RNA。结果 26例HGV/GBV-C与HCV混合感染者PBMC和肝脏中均未检测到HGV/GBV-C负链RNA;而在9份PBMC和15份肝脏标本中检出HGV负链RNA。结论 HGV/GBV-C与HCV混合感染时,在PBMC和肝脏中尚未发现该病毒复制的征据,提示在肝炎病毒混合感染患者中,PBMC和肝脏可能不是HGV/GBV-C的复制场所。
AIM To study the presence of plus-and minus-stranded (replicative intermediate) HGV/GBV-C RNA in the liver, and peripheral blood mononuclear cells (PBMC) of 26 patients with co-infected hepatitis C and G/GBV-Cviruses. METHODS Plus-and minus-stranded HGV/GBV-C RNA and HCV RNA in PBMC and liver were detected with reverse-transcription nested polymerase chain reaction (RT-nPCR) in 32 patients with hepatitis. RESULTS No minus-stranded HGV/GBV-C RNA was found in PBMC and liver sample. Minus-strand HCV RNA was found in 9 cases in PBMC and 15 cases in liver sample. CONCLUSION The liver and PBMC of patients with coinfected hepatitis C and G/GBV-C viruses are not the primary replication sites for HGV/ GBV-C, and the evidence for HGV/GBV-C replication site remains to be clarified.
