癌胚纤维连接蛋白mRNA在大肠癌细胞中的表达

Expression of Oncofetal Fibronectin mRNA in Colon Cancer Cells

目的 检查癌胚纤维连接蛋白在大肠癌中的表达及转化生长因子 β1对其表达的影响。方法 采用逆转录聚合酶链法扩增癌胚纤维连接蛋白 EDA、EDB和 CS1剪切区 ,并对目的片段进行酶切鉴定。结果 EDA、EDB和 CS剪切区在大肠癌细胞中表达阳性 ,EDB、 CS1表达仅局限于癌组织中 ;转化生长因子 β1可部分逆转 EDA、EDB的表达。结论 EDA、EDB和 CS在大肠癌基因诊断和治疗方面具有潜在的应用价值 ,值得进一步研究。转化生长因子β1逆转 EDA、EDB表达可能是该因子抑制大肠癌浸润与转移的机理之一。

Objective To investigate the expression of oncofetal fibronectin mRNA isoforms in colon cancer cells and the effect of transforming growth factor β1(TGFβ1)on its expression.Methods Reverse transcriptase polyperase chain reaction technique was used to amplify the EDA,EDB and ⅢCS1 alternative splicing regions of fibronectin,and the target sequences were identified by specific restrictive enzyme digestion.Results Positive expression of EDA,EDB andⅢCS1 regions were found in colon cancer cells,and the expression of EDB andⅢCS1 Regions were strictly oncogenically regulated.TGFβ1 could partially reverse the expression of EDA and EDB regions.Conclusion The expression of EDA,EDB andⅢCS1regions might play a role in migration and metastasis of colon cancers,while the EDB and ⅢCS1regions might be used as targets for gene diagnosis and therapy.The inhibition of cell mirgration and metastsis induced by TGFβ1 might partially be mediated by the reversion of EDA and EDB expression.
