摘要
目的 :初步探测 MTS1 基因外显子 2缺失及其表达的 P16蛋白阴性在乳腺癌发生发展中的作用。方法 :采用多重PCR扩增 MTS1 基因外显子 2 ,免疫组化染色 (SP法 )检测 P16蛋白表达。结果 :本组乳腺癌中 MTS1 基因外显子 2的纯合子缺失频率为 35 .8% (19/ 5 3)。蛋白阴性占 19.4% (7/ 36 )。三种类型的乳腺癌中 MTS1 基因变异及 P16蛋白阴性频率有明显差异 ,淋巴转移组的基因变异及蛋白阴性频率极显著高于未转移组 (P <0 .0 1)。结论 :MTS1 基因外显子 2变异及 P16蛋白阴性与乳腺癌的病理组织分型相关 ,与乳腺癌的恶性进程密切相关。
Objective:To detect the role of deletion of MTS-1 gene exon2 and loss of P16 protein in breast carcinomas.Methods:MTS-1 gene exon2 and P16 protein were detected by Mutiplex PCR and SP immunohistochemical technique respectively.Results:The deletion rate of MTS-1 gene exon2 was 35.8%(19/53).The negative rate of P16 protein was 19.4%(7/36).There was obvious difference in MTS-1 gene exon 2 deletion and in negative expression of P16 protein among three types of breast carcinomas.And there was significant difference in that between metastatic and non metastatic breast carcinoma of lymph node(P<0.01).Conclution:The deletion of MTS-1 gene and loss expression of P16 protein were associated with the type of breast carcinoma,and were associated with the progression of breast carcinoma.=
