摘要 目的 :研究禁食对大鼠体重及外周脂肪湿重的影响 ,探讨禁食减脂减重与脂肪细胞凋亡的关系。方法 :禁食 48h,用DNA提取物凝胶电泳证实脂肪细胞凋亡的存在 ,逆转录 (RT) - PCR探讨 Bcl- 2与 Bax基因的表达。结果 :1禁食能明显降低大鼠体重 ,组间比较具有显著的统计学意义 (P<0 .0 0 1) ;2禁食能减少外周脂肪组织湿重 ,但统计学意义不显著 (P>0 .0 5 ) ;3DNA提取物凝胶电泳结果证实脂肪细胞发生了凋亡 ;4外周脂肪组织中 Bcl- 2基因表达减少 ,Bax表达增加 ;5生殖器周脂肪组织对细胞凋亡的敏感性高于腹膜后。结论 :降低 Bcl- 2与 Bax的比值 ,诱导脂肪细胞凋亡可能是禁食减脂减重的重要机制。 Objective:To study the influence of fasting on the body and adipose tissue weight of rats and the relation between fasting and white adipocyte apoptosis. Methods:Adipocyte apoptosis and the expression of Bcl 2 and Bax gene were detected by DNA Ladder and RT PCR in male SD rate fasted in 48 hours. Results:Fasting significantly reduced body weight ( P <0.001),but slightly decreased the weight of adipose tissue ( P >0.05). Adipocyte apoptosis was clearly observed by ladder pattern.Bcl 2 gene expression decreased while Bax increased in fasted adipose tissue. And the epididymal adipocyte was more sensitive to apoptosis than retroperitoneal adipocyte. Conclusion:The decreased Bcl 2 gene expression and increased Bax gene expression may be an important mechanism of adipocyte apoptosis induced by fasting.
