HBsAg核酸疫苗诱导H—2^b小鼠体液免疫应答的初步研究 被引量:4

摘要 目的 :观察 adw和 adr亚型 HBV Pre S2 +S核酸疫苗单一或联合 HBc Ag核酸疫苗诱导的小鼠体液免疫反应。方法 :C5 7BL / 6小鼠 2 3只随机分为 4组 :p JW430 3组 (P组 )、p JW430 3/ S2 +S(adw亚型 )组 (W组 )、p JW430 3/ S2 +S(adr亚型 )组 (R组 )、p JW430 3/ S2 +S(adr) +p JW430 3/ HBc组 (R+C组 )。免疫方法为每只小鼠每次肌注相应质粒 DNA10 0 μg(10 0 μl)。免疫程序为 0、2、4周。于第 0、2、4、6、8周采集小鼠血清 ,用 EL ISA法检测血清抗 - HBs和抗 - HBc。结果 :于第 3次免疫后 4周 W组、R组、R+C组小鼠血清全部检出抗 - HBs,抗体含量随免疫次数增多而增加 ,抗 - HBs浓度最高达 2 2 32 9IU / L ,与 P组 (对照质粒组 )相比差异有显著性 (P<0 .0 1) ,但各核酸疫苗组之间差异无显著性 (P>0 .5 )。抗 - HBc在第 1次免疫后 2周即在 R+C组全部出现 ,抗 - HBs和抗 - HBc在 P组始终未检出。结论 :adw和 adr亚型 HBV Pre S2 +S核酸疫苗和 HBc核酸疫苗能诱导 H- 2 b小鼠产生较强的体液免疫反应 ,联合 HBc核酸疫苗有助于抗 - Objective:To observe the humoral immune response induced by the nucleic acid vaccines encoding HBV PreS2+S of subtypes adw and adr and encoding HBcAg. Methods:Twenty three C57BL/6(H 2 b)mice were randomly divided into four groups: Group pJW4303(Group P), Group pJW4303/S2+S(adw) (Group W), Group pJW4303/S2+S(adr) (Group R) and Group pJW4303/S2+S(adr)+pJW4303/HBc(Group R+C). The immunization method is that the corresponding 100 μg(100 μl) of plasmid DNA were injected (i.m) into the quadriceps femoris muscles of each mouse at 0,2,4 wk. At 0,2,4,6,8 wk the mouse sera were collected and measured for anti HBs and anti HBc antibody by ELISA. Results:After the second boost, anti HBs antibody were detected in all mice of group W, group R, and group R+C. Antibody levels increased with boost injection of nucleic acid vaccine, the highest one being 22 329 IU/L. There were most significant differences among W,R,R+C groups compared with Group P( P<0.01 ), but no significant difference observed among W,R,R+C groups ( P >0.5). After the first injection of nucleic acid vaccine of pJW4303/HBc, anti HBc antibody appeared in all mice of group R+C. Neither anti HBs nor anti HBc was detected in group P. Conclusions: Nucleic acid vaccines encoding HBV PreS2+S of subtypes adw and adr and encoding HBcAg indeed induce a strong humoral immune responses. Coimmunization with HBcAg nucleic acid vaccine would facilitate the early production of anti HBs antibody.
