摘要
目的 :探讨辐射后过氧化损伤及VA、VE抗自由基损伤作用 ,为辐射后过氧化损伤的防护提供科学依据。方法 :实验设阴性对照、阳性对照 (辐射 )、VA、VE及VA +VE联合用药 5组。在辐射前每天分别以VA 10mg·kg- 1和VE 30mg·kg- 1给相应组大鼠灌胃。 6d后 ,除阴性对照组外 ,其余大鼠均接受 6Gy 60 Co照射 ,照射后 3d宰杀取血作生化分析。结果 :阳性对照组大鼠出现明显中毒症状。VA、VE和VA +VE组MDA含量分别为 ( 12 8.86± 56.2 7) μmol·gpr- 1、( 110 .84± 33.89) μmol·gpr- 1和 ( 149.94± 32 .82 ) μmol·gpr- 1,显著低于阳性对照组的 ( 2 0 1.2 5±37.0 1) μmol·gpr- 1(P <0 .0 5)。VA、VE和VA +VE组SOD活性分别为 ( 1.35± 0 .79)U·gpr- 1、( 1.66± 0 .61)U·gpr- 1和 ( 1.69± 0 .71)U·gpr- 1,也显著低于阳性对照组的 ( 2 .95± 0 .52 )U·gpr- 1(P <0 .0 5) ;VE组全血CAT活性为 ( 1.4 3± 0 .2 9)U·ml- 1.sec- 1,高于阳性对照组的 ( 1.18± 0 .2 2 )U·ml- 1·sec- 1(P <0 .0 5)。实验组给予VA、VE或VA +VE后 ,血清含量均有所增加。结论 :VA、VE能减弱辐射导致的自由基损伤 ,VA。
Objective:To study the free radical injury of radiation and antioxidative effect of VA and VE, so as to provide scientific basis for protection of peroxidative damage after radiation.Methods:SD rats were divided into 5 groups:negative control,positive control (radiation), VA,VE and VA+VE combined groups.Rats of VA,VE and combined groups were given VA 10 mg·kg -1 or VE 30 mg·kg -1 or both per day. Animals except negative control, were irradiated by 60 CO(6 Gy) 6 days later. All rats were killed and blood samples were collected for biochemical analysis on the 3rd d after radiation.Results:There were obvious toxic symptoms in the positive control group. The levels of serum MDA in VA, VE and VA+VE groups were (128.86±56.27) μmol·g pr -1 , (110.84±33.89) μmol·g pr -1 and (149.94±32.82) μmol·g pr -1 respectively, significantly lower than (201.25±37.0) μmol·g pr -1 in positive control (P< 0.05). The activities of serum SOD were (1.35±0.79) U·g pr -1 ,(1.66±0.61) U·g pr -1 and (1.69±0.71) U·g pr -1 respectively, also significantly lower than (2.95±0.52) U·g pr -1 in positive control (P<0.05). The activity of whole blood CAT in VE group was (1.43±0.29) U· ml -1 ·sec -1 , significantly higher than that of positive control 〔 (1.18±0.22) U·ml -1 ·sec -1 (P<0.05)〕. The levels of serum VA and VE were increased after giving VA or VE. Conclusion: VA,VE could reduce radiation induced-free radical injury. VA combined with VE may also have certain antioxidative effect.
