摘要
目的 探讨 D2 S140基因多态性与 2型糖尿病的关系。方法 运用聚合酶链反应 (PCR)技术 ,结合短串联重复序列 (STR)多态性标记的方法 ,检测了 10 3例 2型糖尿病患者和 10 5例正常对照者的 D2 S140等位基因及基因型。结果 发现 7种等位基因。等位基因 2与 2型糖尿病呈显著性正相关 (RR=13.76 ,P<0 .0 0 1) ,等位基因 6与 2型糖尿病呈显著性负相关 (RR=0 .2 5 ,P<0 .0 0 1)。多因素非条件 L ogistic回归分析表明 ,D2 S140等位基因 2是 2型糖尿病的独立危险因素 ,其相对危险度明显高于环境因素。结论 D2 S140基因多态性与中国人
Objective To determine the relationship between the polymorphism of D2S140 gene and type 2 diabetes mellitus.Methods D2S140 region polymorphism were analyzed with the polymerase chain reaction and short tandem repeat methods in 103 subjects with type 2 diabetes mellitus and 105 nondiabetic subjects.All subjects were biologically unrelated Chinese without a family history of DM.Results There were 7 alleles ranging from 151bp to 163bp in all subjects.The frequencies were 0.01 to 0.37,2 alleles were more common in diabetic patients than in nondiabetic subjects (RR=16.04, P <0.001) and 6 alleles were less frequent in diabetic patients than in nondiabetic subjects (RR=0.23, P <0.001).Logistic regression analysis showed that the 2 alleles of D2S140 gene are main risk factors for type 2 diabetes mellitus.Conclusion Polymorphic microastellite repeat marker at the D2S140 gene locus is positively associated with type 2 diabetes mellitus in Chinese.
