摘要
目的 观察谷氨酸脱羧酶 (GAD) 67mRNA表达在慢性癫痫演变过程中的变化 ,并探讨其作用。方法 采用寡核苷酸探针原位组织杂交技术检测慢性癫痫演变过程中各期皮层、海马GAD67mRNA的表达水平。结果 慢性癫痫演变过程中GAD67mRNA阳性细胞数略有减少 ,但与对照组比较无显著性差异。而继续存活的GAD67mRNA阳性细胞平均吸光度明显增加 ,与对照组比较有显著性差异 (P <0 .0 1)。结论 继续存活的GAD67mRNA阳性细胞GAD67mRNA表达水平的增高可能是机体的一种内源性抗痫表现。
Objective To observe gene expression of GAD 67 mRNA in the brain of rats with chronic epilepsy induced by pentylenetetrazol, and to explicate the possible role of GAD 67 mRNA in the pathogenesis of epilepsy. Methods A model of chronic epilepsy rat was established by PTZ. Cortical and hippocampal GAD 67 mRNA in rats were detected at different time by in situ hybridization. Results The number of GAD 67 mRNA positive cells decrease in kindled rats, but not statistically significant. The average light density was higher in both cortex and hippocampus of kindled rats than in normal rats ( P < 0.01 ). Conclusion The increase of expression of GAD 67 mRNA in survival neurons may be one of endogenous anti epileptic mechanism.
