摘要
目的 动态观察豚鼠关节软骨降解的形态学改变 ,监测其血液和软骨生化指标 ,探讨骨性关节炎早期病变机制。方法 选用Hartley雌性豚鼠 30只 ,分为出生后 1、3、7月龄 ,每组各 10只。观察豚鼠膝关节软骨降解情况 ,测定其血清蛋白多糖和雌二醇水平 ,取胫骨平台软骨测定羟脯氨酸、糖醛酸含量及含水率。结果 3个月组关节软骨即有严重降解。豚鼠的雌二醇水平和软骨含水率随年龄的增长而升高。而血清蛋白多糖和软骨糖醛酸含量随年龄的增长而下降 ,各组间差异非常显著 (P <0 0 1)。软骨羟脯氨酸含量各时间段未见显著差别。结论 原发性骨性关节炎血清及软骨生化指标随年龄增高改变明显。雌激素代谢可能与原发性骨关节炎的发病机制有关。
Objective To investigate the biochemical changes o f primary osteoarthritis of the knee joi nt of guinea pigs and study its initial pathogenesis.Methods 30 female Hartley guinea pigs were divid ed into postnatal 1,3 and 7 months grou ps with 10 animals in each group.The deg eneration of knee joint cartilage was ob served.The proteoglycan(PG)and estradiol (ET)in serum were measured and the conte nts of hydroxyproline(HOP),uronic acid(U A)and water ratio(WR)in articular cartil age were determined.Results Remarkable degeneration of knee joint ca rtilage was seen in guinea pigs of 3 mon ths old.The level of serum ET and cartil age WR increased with age,while the valu e of serum PG and cartilage UA decreased with age.The conent of HOP had no signi ficant difference among 3 groups.Conclusion Biochemical features of primary osteoart hritis in serum and cartilage changed si gnificantly with age.Estradiol metabolis m may attribute to the pathogenesis of p rimary osteoarthritis.
