摘要
用显微分光光度计方法研究了食管癌高发区林县粮食中分离的优势污染菌互隔交链孢霉提取物交链孢酚单甲醚(AME)对人胚肺成纤维细胞(2BS株)的DNA含量和细胞周期的影响。结果发现:AME作用人胚肺2BS细胞两个倍增时间后,随着AME浓度的加大,G_1期细胞峰(2C细胞)降低,出现了较高的G_2+M期细胞峰(4C细胞),S期细胞增加,分裂指数降低.并出现了一些非整倍体的细胞,这些改变与对照组相比较有显著性差异。说明AME可以抑制人胚肺2BS细胞的周期,使S期细胞受阻,堆积和损伤,导致非整倍体细胞出现,这些将有可能成为细胞突变的遗传基础。从而为互隔交链孢霉与林县人食管癌的发生可能有一定关系的说法提供了实验佐证。
We studied the effects of alternariol monmethyl ether (AME), a product of alternaria isolatedfrom the grain in the area of high incidence of esophageal cancer. Linxian,on the DNA contents of thefetal lung 2BS cells and the cell cycles with cytophotometry. The DNA contents of the 2BS cells weredetermined after the 2BS cells had been treated for two doubling time. The results showed that with theincrease of the AME concentration the G_1 phase cell (2C cell) peak decreased,thc G2+M phase cell(4C cell) peak increased,S phase cells significantly increased,MI decreased and some aneuploid cells e-merged. These changes were significantly different from those of the control (P<0. 01), indicatingthat AME might inhibit the cell cycles, make S phase cells blocked, accumulated and damaged, andcause the aneuploid cells to emerge,which could be the hereditary foundation of cell mutation. Theabove results provided some experiement data for the hypothesis that alternaria alternata might be oneof the etiological factors of human esophageal cancer in Linxian.
