摘要
目的 通过大鼠脑缺血再灌注模型 ,观察缺血 1 5h后 ,再灌注 1 2、2 4、4 8和 72h皮层、尾壳核和海马细胞凋亡的变化。方法 采用线栓法制成大鼠左侧大脑中动脉缺血再灌注模型 (n =2 1 ) ,应用末端转移酶介导的dUTP -DIG缺口末端标记法 (TUNEL) ,标记凋亡细胞断裂的DNA。结果 脑缺血再灌注后 1 2h,左侧大脑中动脉缺血区皮层和尾壳核出现大量凋亡细胞 ,于再灌注后 2 4~ 4 8h达高峰。
Objective To investigate the neuronal apoptosis in the area of cortex,caudoputamen and hippocampus after 1.5 h ischemia and reperfusionat different time intervals (12,24,48 and 72 h).Methods The focal cerebral ischemia-reperfusion models of rats (n=21) were made by thread embolism of left middle cerebral artery (LMCA).DNA fragmentations of apoptotic cells were detected with terminal deoxynucletidyl transferase (TdT)-mediated dUTP-DIG nick end-labeling (TUNEL).Results The apoptotic cells exhibiting DNA fragmentation increased in the cortex and caudoputamen of core ischemic area of LMCA at 12 h ischemia-reperfusion and peaked at 24 -48 h after reperfusion.Conclusions Focal ischemia-reperfusion can induce apoptosis in early stage.
