摘要
用高压匀浆机对重组大肠杆菌细胞进行破碎以释放重组人白细胞介素-6(rhIL-6)包含体,系统研究了高压匀浆过程中物理和化学因素的影响,包括匀浆次数,操作压力,料液的pH 及悬浮体系的影响。从而选择操作压力为80MPa,悬浮体系为去离子水,经3次破碎,表达量为20%的菌体可获得粗包含体的纯度约为40%。此外,本文还对该高压匀浆破碎过程进行了动力学分析。
The disruption of recombinant E.Coli cells by a high-pressure homogenizer was investigated for the extraction of rhIL-6 inclusion bodies. A systematic study was carried out on the influence of physical and chemical factors including homogenizer passes, operating pressure and suspension system during homogenization processes. Under exprimental conditions: 80Mpa, deionized water suspending systen, 3 passes through the homogenizer, the original pellet of inclusion bodies contained about 40% rhIL-6 can be achieved from recombinant E.Coli which rhIL-6 accumulation levels is of 20% total cell protein. Meanwhile , kinetic studies of this disruption process had been made, a kinetic model was uesd and kinetic parameters were obtained under different conditions.
