EB病毒BHRF1基因表达对鼻咽癌细胞增殖周期影响的研究

摘要 目的:了解EB病毒编码BHRF1基因表达对鼻咽癌细胞增殖周期的影响。方法:本研究通过构建BHRF1高表达载体并将其转入鼻咽癌细胞株CNE2细胞中,检测转染后稳定表达BHRF1癌细胞的增殖细胞核抗原(proliferatingcelnuclearantigen,PCNA)表达和喜树碱作用前后细胞周期分布的改变。结果:BHRF1的表达可促使癌细胞的PCNA表达下降,G1期细胞增多,S期细胞减少;而在喜树碱作用后,BHRF1-CNE2细胞的凋亡率明显小于对照组,其G1期细胞数下降,S期细胞数升高。结论:BHRF1基因的表达可通过调节细胞周期分布、抑制细胞凋亡的发生以增强细胞的生存能力。 Objective: To investigate the effect of expression of EB virus gene BHRF1 on the redistribution of cell cycle of the cells of nasopharyngeal carcinoma. Methods: BHRF1 high expressing vectors were constructed and transfered into the cell line of nasopharyngeal carcinoma, CNE2. Then proliferating cell nuclear antigen (PCNA) and the redistribution of cell cycle in the cells after camptothecin treatment were tested. Results: BHRF1 expression could reduce the expression of PCNA, increase the cells in G 1 phase and decrease the cells in S phase. After camptothecin treatment. The apoptotic rate of BHRF1-CNE2 cells was lower than control groups, and its cells in G 1 phase decreased and in S phase increased. Conclusion: The expression of BHRF1 gene can regulate the distribution of cell cycle, enhance cell survival by inhibiting its apoptosis, and may be related to the developement of nasopharyngeal carcinoma.
