摘要
目的 建立一种能动态显示活体内肿瘤细胞生长、新生血管形成及靶基因表达水平等的新型动物模型。方法 以海洋水母中分离到的绿色荧光蛋白 (GFP)作为报告基因 ,将 1× 10 3~ 1× 10 4 持续、稳定、高水平表达GFP的肿瘤细胞移植到鼠皮窗内 ;或以 1× 10 8~ 1× 10 9GFP腺病毒感染皮窗肿瘤 ;或将转导了热诱导型表达质粒的肿瘤细胞移植到鼠皮窗内 ,隔天用荧光显微镜进行观察。结果 移植到鼠皮窗内的肿瘤细胞 3~ 7d时可见肿瘤细胞围绕大血管生长及大血管出芽形成新生血管 ,腺病毒的感染效率及靶基因的表达水平可通过GFP表达确定。结论 新型肿瘤皮窗模型可显示肿瘤细胞生长及新生血管形成过程 。
Objective To establish a new rodent tumor model for observing tumor cell growth and angiogenesis in the early stage of tumor formation in vivo, and evaluating gene expression in tumor and efficiency of anti cancer therapies. Methods The plasmid containing GFP cDNA was transfected into tumor cells by liposome or electroporation. The tumor cells with stable and high level expression of GFP protein were selected by using G418 and subcloning, and then implanted into mice or rat dorsal skin window chamber. The tumor cell growth, angiogenesis and pattern of gene expression either delivered by adenovirus or instinct in tumor were observed under a microscope. Rresults Both tumor cell growth and angiogenesis could be observed simultanously, noninvasively and serially with unprecedented clarity. The expression efficiency and kinetics of gene delivered by adenovirus or transcriotional activity of interested genes in the tumor could also be monitored. Conclusion The novel rodent tumor model provides a powerful and versatile tool for observing tumor cell growth and angiogenesis in vivo, and evaluating the efficiency of anti cancer therapy.
