摘要
目的 :观察不同缺铁饲养期对大鼠耳蜗肌动蛋白含量及分布的影响 ,探讨铁缺乏致聋的发病机制。方法 :应用 SDS-PAGE蛋白电泳、双波长薄层扫描分析、免疫组织化学以及听性脑干诱发电位检测 ,观察比较铁缺乏 8周、12周的大鼠及正常大鼠耳蜗肌动蛋白含量与分布的变化。 结果 :与正常对照大鼠比较 ,铁缺乏引起的感音神经性聋大鼠耳蜗肌动蛋白相对含量明显下降 (P<0 .0 1) ,免疫组化反应减弱 ;但不同缺铁期间耳聋大鼠耳蜗肌动蛋白变化比较无显著性差异 (P>0 .0 5 )。 结论 :耳蜗内肌动蛋白相对含量减少和 (或 )免疫组化反应减弱与铁缺乏致聋存在密切关系 ;缺铁时间长短对大鼠耳蜗肌动蛋白含量及免疫组化反应无明显影响。
Objective: To observe the content and distribution of cochlear actin in the different periods iron deficient(ID) rats.Methods: Actin of cochlear membranous tissue was separated and determined by SDS PAGE and Western blot; the relative content and distribution of cochlear actin in 8 weeks, 12 weeks iron deficient rats and normal rats were studied by TLC densitometric determination and immunohistochemistry. Results: All rats in different groups suffered from hearing loss had reduction of relative content and immunohistical reaction of chochlear actin,whereas there were no significant differences among them.Conclusion: The reduction of actin in chochlea is one of the important pathological basis for sensorineural hearing loss in iron deficient rats; the periods of iron deficiency have no significant influence on the relative content and distribution of cochlear actin in rats.
