摘要
目的 :为进一步提高产乳糖酶酵母 Kluyveromyces fragilis(K.fragilis)株的酶产量 ,对该菌株的培养产酶发酵条件进行优化试验。方法 :采用增殖和诱导二步培养法培养酵母 K.fragilis株 ,并采用单因素试验方法分析碳源、氮源对酵母诱导产酶的影响。结果 :确定了 K .fragilis酵母株诱导培养的较佳配方 ,当乳糖浓度为 1 2 0 g/ L,醋酸铵浓度为 5 g/ L,p H为 6 .7时 ,单位酶产量达1 2 2 .4IU/ ml培养液。结论 :该菌株生长产酶情况良好 。
Objective: To study the optimal condition on lactase production by yeast Kluyveromyces fragilis(K.fragilis) to increase the enzyme productivity.Methods: With separated proliferation and induction culture,the effects of carbon source and nitrogen source on yeast proliferation and lactase induction were studied by the method of monofactorial analysis.Results: The optimum medium for the productivity of lactase by K.fragilis was determined.Maximal enzyme activity (122.4 IU/ml) was reached when the concentration of lactose was 12%,ammonium acetate was 0.5% and pH 6.7 .Conclusion: The results of this experiment demonstrate that this strain will be good for industrial production.
