摘要
对溶葡球菌酶生产菌BacillusSubtilis-0041的发酵条件和工艺作了进一步研究和改进,该菌的摇瓶产酶可达550u/ml,在5L自动发酵罐上达680u/ml,且发酵周期大大缩短,仅5~6小时,发酵产率比原工艺提高一倍以上。滚工艺全部使用国产常用原料,为产业化生产提供便利。在研究中发现溶解氧对培养过程非常重要;pH对产酶没有明显影响;发酵中期流加葡萄糖可抑制蛋白酶对产物酶的降解作用。
n this experiment,the fermentation conditions have been widely investigated
and improved inLysostaphin preducer B. subtilis 0044. In new condition,through 5-6hr.B.
subtilis 0044 can produceLysostaphin 550u/ml in flask and 580u/ml in 5L fermenter.The
Lysostaphin yield has increased by100% compared with original condition, The fermentation
period has shortened.In the new technology,all components are from China it is suitable for
industrial scale production.Through experirnent,itwas discovered that diss,olved oxygen is
very improtant in Lysostaphin fermenta-tion,But pH is not adistinct effector.In the process of
fermen tation, The addit ion of glucose or organic nitrogen can prevent decom positionof
Lysostaphin by proteinase.
