红细胞作载体的谷胱甘肽过氧化物酶用于脑缺血-再灌注的研究 被引量:10

摘要 目的和方法:采用红细胞(Erythrocytes,RBC)作为谷胱甘肽过氧化物酶(glutathioneperoxidase,GSH-Px)的载体,将包载GSH-Px的RBC(RBC-GSH-Px)用于兔脑缺血-再灌注(ischemia-reperfusion,I-R)研究。结果:包入采用低渗透析法,渗透压达到132mOsm。采用51Cr标记法测定的兔RBC的循环半寿期为(134±15)d(n=6),RBC-GSH-Px的半寿期为(73±25)d(n=5)。2,3,5—氯化三苯基四氮唑(TTC)染色结果表明,RBC-GSH-Px对I-R损伤的脑组织有明显的保护作用。RBC-GSH-Px对一氧化氮(nitricoxide,NO)浓度的升高有抑制作用。 AIM and METHODS:The practicability using red blood cells(RBC)as the carrier of glutathione peroxidase(GSH-Px) was studied in this paper.GSH-Px was encapsulated into rabbit erythrocytes (RBC-GSH-Px),and was used in the study of cerebral ischemia-reperfusion(I-R) The effect of RBC-GSGH-Px on cerebral I-R was observed.RESULTS:The method of hypotonic dialysis was used in preparing RBC-GSH-Px,the osmotic pressure reached 132 mOsm.The circulation half-life (Τ1/2) of RBC-GSH-Px was determined in rabbits by 51 Cr radiolabel.Τ1/2=(13 4±1 5)d( n =6) for RBC,and Τ1/2=(7 3±2 5)d( n =5)d for RBC-GSH-Px.The increase of nitric oxide (NO) was inhibited by the administration of RBC-GSH-Px Staining with TTC showed that RBC-GSH-Px effectively protected cerebrum from I-R damage.CONCLUSION:Using RBC as the carrier of GSH-Px is practicable,it may be effective in the treatment of some brain diseases.
