Abstract Macrophages synthesize nitric oxide (NO) through L-arginine-NO pathway when nitric oxide synthase is induced by LPS and cytokine. In inflammatory tissues, the major site of nitric oxide synthase induction is macrophages. NO released from macrophages causes relaxation of myocardium and vascular smooth muscle in vivo and in vitro . These results suggest ed that NO released from activated macrophages plays an important role in vasodilation of endotoxemia and septic shock. In patients with endotoxemia or septic shock, adminitration of L-arginine analogues and glucocorticoids may be effective in inhibiting ON synthesis and preventing hypotension.
