钠尿肽抑制SD乳鼠心肌细胞增殖 被引量:4

Inhibitory effect of natriuretic peptides on proliferation of cardiac myocytes in neonatal rats

摘要 目的 比较心房钠尿肽 (ANP)、血管钠肽 (VNP)及C-型钠尿肽 (CNP)抑制心肌细胞增殖的效应 .方法 体外培养 SD乳鼠心肌细胞 ,用蛋白激酶 C激动剂佛波酯 (PMA)刺激心肌细胞增殖 ,以总蛋白含量和噻唑蓝 (MTT)比色吸光度值为指标 ,观察 3种钠尿肽抑制心肌细胞增殖的效应 .结果 PMA(10 - 9~ 10 - 7mol· L- 1 )致使心肌细胞的总蛋白含量和MTT吸光度值显著升高 (P<0 .0 5 ) ,ANP(10 - 8~ 10 - 6 mol·L- 1 ) ,VNP和 CNP(10 - 7~ 10 - 6 mol· L- 1 )均可显著降低PMA(10 - 8mol· L- 1 )刺激的心肌细胞总蛋白含量和 MTT吸光度值 (P<0 .0 5 ) .结论  PMA对心肌细胞有促增殖作用 ,ANP,VNP和 CNP均可抑制 PMA刺激的心肌细胞增殖 ,抑制效应以 AIM To compare the inhibitory effects of vasonatrin peptide (VNP), atrial natriuretic peptide (ANP) and C type natriuretic peptide (CNP) on the proliferation of cardiac myocytes. METHODS Total protein and optical density (OD) value of MTT were detected in cultured neonatal rat cardiac myocytes induced by phorbol 12 myristate 13 acetate (PMA). RESULTS PMA(10 -9 ~10 -7 mol·L -1 )significantly increased the total protein and OD value of MTT in cardiac myocytes ( P <0.05) and ANP (10 -8 ~10 -6 mol·L -1 ), VNP and CNP (10 -7 ~10 -6 mol·L -1 ) significantly decreased them ( P <0.05) induced by PMA(10 -8 mol·L -1 ) in a dose dependent way respectively. CONCLUSION These data suggest that PMA stimulates the proliferation of cardiac myocytes while ANP, VNP and CNP inhibit PMA induced proliferation of cardiac myocytes in rats and ANP has the strongest inhibitory effects.
