摘要
目的 在裸鼠体内建立人骨肉瘤自发性高肺转移模型 ,为探讨骨肉瘤肺转移机制和抗转移治疗提供尽可能与临床条件相近的实验工具 .方法 将裸小鼠腹腔筛选的人骨肉瘤高转移细胞亚群经裸鼠间传代 ,组织块原位植入胫骨骨髓腔 ,待裸鼠发生转移时 ,皮下扩增肺转移灶 ,再次原位移植 .如此循环 3个周期 .观察各代肿瘤细胞致瘤率、脏器转移及染色体数目变化情况 .结果 各代肿瘤细胞致瘤率均为 10 0 % ,肺转移率稳定在 80 %以上 ,转移途径单一 ;体内增殖能力及染色体数目稳定 .结论 建立了人成骨肉瘤自发性高肺转移模型 。
AIM To establish a model of human osteosarcoma with spontaneous highly pulmonary metastasis in nude mice and to study its biological characteristics. METHODS Histologically intact tumor pieces from solid subcutaneous tumors gained from highly metastatic subpopulation were inoculated into the tibia of nude mice, the representatives of pulmonary metastases were obtained, expanded subscutaneously and orthotopically inoculated again. The processes were repeated three times and changes were observed in intratibial tumor growth, pulmonary metastatic rate and the chromosome numbers in every passage in vivo . RESULTS All mice developed intratibial tumors and the pulmonary metastatic rates were higher than 80%. The metastatic paths were single and the changes of the chromosome numbers were stable. CONCLUSION A model of human osteosarcoma with spontaneous highly pulmonary metastasis is established in nude mice, and its biological characteristics are stable during passage in vivo .
