摘要
目的 考察低分子肝素肺靶向微球缓释性、稳定性和靶向性。方法 用乳化法制备微球 ,正交试验设计考察影响制备工艺的因素 ,体外释药试验研究其缓释性 ,以外观形态、粒度分布、包封率、释药性等的变化考察其稳定性 ,以及微球在家兔的体内分布。结果 所制备微球粒径在 5 2 5 μm者占 85 .5 % ,包封率为 87% ,并有良好的缓释性、靶向性和稳定性。结论 该法制备的微球稳定 ,可用于肺靶向研究。
AIM To test gelatin microspheres of low molecular weight heparin(LMWH-GMS) release behavior; lung target behavior and stability. METHODS The microspheres were prepared with the emulsifing method and some factors effecting the technology were analysed with the orthogonal test design. The sustained release of LMWH-GMS was studied in vitro and preliminary stability of LMWH-GMS was investigated by the changes of conformation, diameter distribution, encapsulated ratio and the drug delivery of LMWH-GMS. The lung target was tested preliminarily by the distribution in different tissues in rabbits. RESULTS LMWH-GMS had a significantly sustained release behavior, a good lung target and stability. CONCLUSION LMWH-GMS can be used for study of lung target.
