摘要
目的 探讨缺血预处理 (IP)对大鼠移植肝缺血再灌注损伤的保护作用及可能机制。方法 制备三袖套大鼠同种异体原位肝移植 (OLT)模型 ,将其分为OLT对照组和IP组 ,用自动生化分析仪检测血中ALT、AST和LDH ,用高压液相色谱仪测定移植肝组织中腺苷酸含量的变化。结果 IP后血中ALT、AST和LDH均较OLT组显著降低 ;肝细胞浊肿、变性 ,肝窦变窄 ,红细胞聚集及大量的中性白细胞和单核细胞附壁 ,局部小叶结构的破坏等变化也较OLT组为轻 ;移植肝组织中ATP、ADP和能荷 (EC)较OLT组明显升高。结论 IP可以维持肝组织中较高浓度的ATP ,这可能是减轻移植肝缺血再灌注损伤及恢复肝细胞功能的机制。
Objective To evaluate the effe cts of ischemic preconditioning(IP)on ischemia/reperfusion(I/R)injury in rats with orthotopic liver transplantation(OLT)and its mechanism.Methods The orthoto pic liver transplantation in the rat was made by three cuffs.OLT rats were divided into two groups:OLT control(OLTC)and IP.The hepatic function and hepatic adenyl i c acids were determined by autochemical analysor and high performance liquid chr omatography respectively.Results The serum values of ALT,AST and LDH in IP rats significantly decreased compared with OLTC.The pathologic changes including edema,degeneration, narrowed sinosoids and quantity of neutrophils in grafts we re more markedly alleviated in IP rats than those in OLTC.The concentrations of ATP a nd ADP,and energy charge(EC)in grafts were higher in IP rats than those in OLTC rats.Conclusion The results demonstrate that the higher concentration of AT P in the graft is maintained in OLT rats with IP,which may be responsible for al leviating I/R injury of liver graft and renewing hepatic function.
