摘要 采用超临界CO_2流体萃取(SFE)金毛狗脊中磷脂类成分,通过均匀设计法考察了SFE的萃取条件,确定萃取压力为31.5MPa,温度为46.2℃,改性剂量为0.37L/kg药材,萃取时间设定为2h。然后采用反相高效液相色谱法,以Waters Symmetry C_(18)为固定相,甲醇-1.0%磷酸溶液(90∶10)为流动相,在检测波长为206nm条件下分离测定磷脂酰胆碱的含量为0.198%(RSD4.2%),为金毛狗脊及其制剂的质量控制提供了方法。 A supercritical fluid extraction (SFE) method was used to extract phospholipids in Cibotium barometz. After a homogeneous design experiment, SFE conditions were obtained which were pressure at 31.5 MPa, temperature at 46.2℃and a 0.37 L/kg modifier in supercritical fluid. The sample was extracted for 2 hours. A RP-HPLC method was developed for the determination of phosphatidylcholine. RP-HPLC conditions included a RP-HPLC column packed with Waters Symmetry C18(5 μm), a mobile phase composed of methanol-1 % phosh-poric acid(90:10) and the detection wavelength at 206 nm. The content of phosphatidylcholine was 0.198% (RSD 4.2%), and the method can be a measure of quality control for Cibotium barometz and its preparations.
