用国产单克隆抗体纯化重组干扰素α_(1b)的研究

Study on the purification of recombinant interferon alpha-1b with domestic monoclonal antibody affinify chromatography beads

制备高纯度的重组干扰素α1b的关键技术是单克隆抗体亲和层析。我们过去用英国Celltech公司提供的抗干扰素α单克隆抗体珠进行纯化。为适应大规模生产的需要 ,我们用安科生物高技术公司所建的杂交瘤细胞分泌的干扰素单克隆抗体及其制备的抗体珠进行纯化 ,获得较好的结果 ,纯化IFNα1b原液纯度 (SDS PAGE ,HPLC ,比活 ) ,鼠IgG及单克隆抗体珠的吸附量 ,使用次数等均符合要求 。

The recombinat interferon alpha 1b (IFNα 1b )has been licensed by the Ministry of Public Health as a product for formal production. Its principal purification process is monoclonal antibody affinity chromatography.Formerly the monoclonal antibody (McAb) beads used in our laboratory were obtained from Celltech Company,that gave rise to good rdsults,but it is rather expensive and in short supply.The McAb prepared by AnKe Biologcal and Technology Comanany was performed excellently too,including the results of IFNα 1b purity (SDS PAGE,HPLC,specific activity),residual content of mouse IgG and the amount of IFNα 1b absorbed per ml McAb beads and usage times of McAb,all of them are in confirmation with the regulation for large scale preparation in this study.
