抗TPO单克隆抗体杂交瘤细胞系的建立

The preliminary determination of anti-TPO monoclonal antibody hybridoma cell line

用合成肽TPO作抗原 ,经腹腔免疫Balb c小鼠 ,鼠抗血清效价为 1:10 0 0 ,ELISA间接法测定的P N值为 3。取小鼠脾细胞与骨髓瘤细胞 (SP2 0 )在PEG作用下进行融合 ,细胞融合率达 91%。通过ELISA筛选并经过 3次亚克隆 ,获得了 1株能分泌抗TPO抗体的单克隆杂交瘤细胞株 ,P N值均高于 8。

Bal b/c mice were immunized with TPO antigen from synthesized peptide, their spleen cells were fused with myeloma cells by polyethyleneglyco (PEG)making for hybridomas cell line. The rate of fusion was 91%. By making ELISA screening and subcloning, one hybridoma cell line was established.
