摘要
采用免疫组织化学方法观察了反复听源性惊厥 (AGS)对遗传性癫易感大鼠P77PMC脑内脑源性神经营养因子 (BDNF)蛋白表达的影响。结果发现 ,在未发生AGS的P77PMC大鼠脑内 ,BDNF蛋白的基础表达可见于海马齿状回门区神经元、CA3区锥体细胞及大脑皮质神经元的胞浆内。在经过 30次AGS发作后 ,鼠脑内齿状回门区、CA1~CA3区以及皮质区免疫标记阳性的神经元数目增多 ,标记强度增强。除了胞体 ,在神经元突起上也可见明显的免疫标记。这表明在P77PMC这种遗传性癫易感大鼠模型中 ,非人为伤害因素造成的惊厥反复发作能导致脑内BDNF表达显著增高。
We examined the effects of recurrent audiogenic seizures (AGSs) on BDNF protein expression in the brain of P77PMC rats using immunohistochemistry technique. The basal expression of BDNF protein was seen in the neuronal cytoplasma of the hilar region of the dentate gyrus, the CA3 hippocampal region and the superficial layers of cerebral cortex. After 30 times of AGSs, the expression of BDNF was dramatically upregulated in the dentate hilar region, CA1~CA3 region and cerebral cortex. Cytoplasmic BDNF-immunoreactivity(IR) was markedly increased in both intensity and number of neuronal cell labeled, and the BDNF-IR was also seen in the axons and dendrites of the neurons. These data suggest recurrent AGSs themselves induce upregulation of BDNF expression in P77PMC rats in which the seizures are not induced by any insults to the brain.
