摘要
目的 :观察传代培养的PC12细胞的无丝分裂和不对称分裂 ,探讨其在肿瘤发生、肿瘤细胞多形性和细胞分化中的意义。方法 :传代培养的PC12细胞以Giemsa染色、甲绿 派郎宁的细胞化学和增殖细胞核抗原 (PCNA)的免疫组织化学染色 ,光镜下分别观察。结果 :PC12细胞呈现横缢型和侧凹型等无丝分裂像 ,并且观察到其形态、大小和化学性质的不对称性分裂及细胞脱颖现象。结论 :无丝分裂和不对称分裂是PC12细胞多形性形成的主要原因 ,肿瘤细胞无丝分裂及其不对称性对肿瘤发生及肿瘤细胞的分化具有重要意义。
Aim:To observe the amitosis and asymmetric division in subcultured PC12 cells, and to explore the role of amitosis and asymmetric division in tumorigenesis, tumor cell pleomorphism and differentiation.Methods: Subcultured PC12 cells were stained with Giemsa staining, methyl green pyronin by cytochemistry and proliferating cell nuclear antigen (PCNA) by immunohistochemistry. The stained samples were observed with the light microscope respectively.Results: PC12 cells exhibited amitotic figures including circular shrink, sideways concave, etc.The figures of asymmetric division in shape, size and the cell chemical properties by staining and the cell enucleation were observed.Conclusion: Amitosis and asymmetric division play an important role in PC12 cell pleomorphism. The asymmetric division possesses important significance in tumorigenesis and differentiation.
