摘要
利用体外分离培养人胚成骨样细胞,加入不同剂量胰岛素样生长因子-Ⅱ(IGF-Ⅱ),从时间-效应和剂量-效应两个方面考察IGF-Ⅱ对人胚成骨样细胞增殖与分化的影响。研究结果表明,IGF-Ⅱ的促骨形成效应显著,它对体外培养的人胚成骨样细胞DNA、胶原蛋白和碱性磷酸酶合成均有明显的促进作用,其作用亦呈剂量与效应相依赖的关系,在4个含量组中,10ng/mlIGF-Ⅱ对成骨样细胞的作用最强。IGF-Ⅱ是一种对成骨细胞作用较全面的生长因子。
Biological regulatory mechanism which supports bone cell development within the context of proliferation/differentiation relationship serves as a viable basis for addressing skeletal disorders and provides experiment and theoretic basis for selection and ultilization of growth factors to reconstruct bone defect.Osteoblastlike cells isolated from human calvarium were used in this study.IGFⅡ were investigated by adding in the culture medium with different concentration. The results showed that IGFⅡ stimulated DNA,collagen synthesis and ALP activity of human osteoblastlike cells.Its effects were strong for proliferation and differentiation of osteoblast cells, when 10ng/ml of four different concertrations (0.1,1,10 and 100 ng/ml) were used.IGFⅡ was proved to be important factor in bone regeneration.
