摘要
目的 :探讨诊断超声对大鼠睾丸组织生精细胞P5 3 ,Bcl 2蛋白表达的影响。材料与方法 :32只SD雄性大鼠随机分为 4组 :Ⅰ组 (对照组 ,空照 ) ,Ⅱ组 (照射 10min组 ) ,Ⅲ组 (照射 2 0min组 ) ,Ⅳ组 (照射 30min组 )。应用HP85 0 0彩色多普勒超声诊断仪对大鼠睾丸组织分别进行不同时间的持续照射 ,2 4h后处死动物取睾丸进行免疫组化分析 ,观察P5 3 ,Bcl 2表达情况。结果 :正常对照组睾丸组织P5 3表达率 (2 1 8± 3 49) % ,Bcl 2表达率 (88 0 6± 2 1 34) %。Ⅱ组与Ⅰ组无差异 ,Ⅲ组和Ⅳ组P5 3表达率显著增高 ,Bcl 2表达率显著降低。结论 :超声诊断剂量照射大鼠睾丸组织 10min以上可引起P5 3蛋白高表达 ,Bcl 2低表达 。
Prupose: To investigate the effect of ultrasound exposure of on P53 and Bcl 2 protein expression rat testes. Marterials and Methods: 32 male Sprague Danles rats were randomly selected into four groups according to different exposure time: group Ⅰ (control), group Ⅱ (10 minutes exposure), group Ⅲ (20 minutes), group Ⅳ (30 minutes). HP 8500 color Dopper sonography with 7.5MHz linear phased array transducer were used for radiation of testes. The testes were removed 24 hour after radiation. Experession of P53 and Bcl 2 protein was analyzed with immunohistochemistry. Results: The positive rate of P53 protein in control group was 21.83%±3.49%, that of Bcl 2 was 88.06%±21.34%. There was no difference between control group and group Ⅱ. The higher expression of P53 protein and the lower expression of Bcl 2 were apparent in group Ⅲ and Ⅳ. Conclusion: Continuous ultrasound exposure over 10 minutes may result in higher expression of P53 and lower expression of Bcl 2 in rat testes, which might further induce germ cell apoptosis.
