摘要
目的 :观察褪黑素对体外培养人外周血淋巴细胞的姐妹染色单体交换(SCE)和细胞增殖动力学的影响。方法 :以33.4μg/L的丝裂霉素(MMC)为阳性对照 ,0.5%乙醇为阴性对照 ,用0.01,0.10,1.00mmol/L褪黑素分别预处理细胞后37℃±1℃体外培养72h ,观察褪黑素处理组及丝裂霉素加褪黑素处理组细胞的SCE频率和淋巴细胞增殖指数。结果 :单独用0.01,0.10,1.00mmol/L褪黑素处理组的SCE频率与空白对照组比较 ,未见有统计学意义 ,而丝裂霉素加不同浓度褪黑素处理组的SCE频率与阳性对照丝裂霉素处理组比较差别有显著性 ,呈明显降低。淋巴细胞增殖指数只有1.00mmol/L褪黑素处理组较空白对照组明显降低。结论 :褪黑素无遗传毒性 ,而且在超生理浓度时具有抗诱变作用 。
Objective:To observe the effect of melatonin on frequency of sister chromatid exchange(SCE) and cell proliferation kenitics in lymphocytes from human peripheral blood in vitro. Methods: Cells were cultured in vitro in the presence of 0.01,0.10,1.00mmol/L melatonin ,33.4 μg/L mitomycin C(MMC),0.01,0.10,1.00mmol/L melatonin plus MMC and 0.5% ethanol for 72h at 37℃±1℃.Lymphocytes were examined for proliferation indices,and for the incidence of SCE. Results: The incidence of sister chromatid exchange of lymphocytes which cultured in the presence of 0.01,0.10,1.00mmol/L melatonin compared with negative control exhibited no statistic significance,the SCE of cells treated with melatonin plus mitomycin C compared with positive control was markedly decreased.The proliferation index was significant lower than negative control only in the culture exposed to 1.00mmol/L melatonin. Conclusion: In vitro assay,it seems that melatonin has not only no genotoxic,but also anti-mutagenic effect on lymphocyte,and has some influence on cell proliferation kenetics at supraphysiological concentrations.
