摘要
利用结球生菜叶片外植体进行组织培养,成功地诱导出愈伤组织与体细胞胚,获得了再生植株,在培养过程中随着组织的分化与脱化历程,过氧化物酶同工酶谱的活性增强,酶谱带增多,并大多数集中在“一”极酶带区,而且分化时比脱分化时增强更显著。用细胞分裂素KT与BA诱导分化时均具有相同的过氧化物酶同工酶谱,但加BA时除了器官分化还有胚胎发生,酶带显色更深。用IAA与NAA诱导苗基部发根过程中,过氧化物酶同工酶谱带比分化芽的酶谱带数少,显色更浅。这反映了形态发生与过氧化物酶同工酶有密切的关系。
Calli, embryoids and plantlets have been obtained from leaves ofhead lettuce in vitro The peroxidase isozyme pattern changed with differenti-ation and dedifferentiation of explants, including that the number of peroxidaseisozyme increased mainly in negative pole and the activity of them was of morestrength in dedifferentiation than in differentiation MS medium separaoelyadded 0.2mg/lKT+0.1mg/l LAA and 0.2mg/l BA+0.1mg/l IAA which can pr-omote budding of head lettuce. Although they have the similar peroxidase isoz-yme, their activity was of more strength on MS medium with BA and IAAbecause of producing embyoids while producing buds on it MS medium withIAA and NAA which can promote rooting of head lettuce. The number ofperoxidase isozyme was less in forning roots than in forming buds, so was thecolour of peroxidase isozyme.
