补阳还五汤防止兔模拟经皮冠脉腔内血管成形术后再狭窄的实验研究

Imitative Restenosis after Percutaneous Transluminal Coronary Angioplasty Prevented by Buyang Huanwu Decoction in Rabbits

目的 :观察补阳还五汤 (消栓口服液 )对饲料所致动脉粥样硬化兔模拟经皮冠脉腔内血管成形术 (PTCA)后再狭窄有无防止的作用。方法 :动物随机分中药组 2 4只和对照组 2 0只 (另 4只术中死亡 )。中药组术前 1周至术后 4周喂药 2ml/ (kg·d) (含生药 8 4 g/ml) ;对照组喂同量生理盐水。两组均在腹主动脉用球囊法模拟PTCA。结果 :术后 60天中药组和对照组受损血管内膜增生发生率分别为 2 9 2 %、85 0 % ;内膜增生面积分别为 ( 0 54± 0 2 1)mm2 、( 1 67± 0 75)mm2 ;内膜增生百分比为 ( 18 0± 7 2 ) %、( 56 0±17 1) % ;管腔相对面积比为 ( 0 74± 0 0 3) %、( 0 35± 0 0 7) %。以上差异均有显著性 (P <0 0 1)。含药血清对血管平滑肌细胞培养的3H TdR掺入有显著抑制作用 ,中药组和对照组分别为 ( 4 110 14± 1977 0 1)cpm、( 7960 14± 2 80 2 59)cpm(P <0 0 5)。 结论 :补阳还五汤 (消栓口服液 )对饲料所致动脉粥样硬化兔模拟PTCA后再狭窄有显著防止作用。

Objective: To examine the preventive effect of Buyang Huanwu Decoction (BYHWD) on imitative restenosis after percutaneous transluminal coronary angioplasty (PTCA) in diet induced atherosclerotic rabbits.Methods: Diet induced atherosclerotic rabbits were randomly allocated into group 1 (n=24, fed with BYHWD one week before and 4 weeks after the operation, 2ml/kg per day, containing herbal drug 8.4 g/ml) and group 2 (n=20, as control). Balloon angioplasty was performed in the abdominal aorta to mimic coronary angioplasty. Results: Sixty days after operation: (1)Incidence of hyperplasia of the intima was 7/24 (29.2%) in group 1 vs 17/20 (85.0%) in group 2, P<0 01; (2) Comparison of area of hyperplastic intima was (0 54±0 21)mm 2 vs (1 67±0 75) mm 2, P<0 01;(3)Area of hyperplastic intima/area of lumen (limited by the internal elastic membrane was (18 0±7 2)% vs (56 0±17 1)%, P<0 01; (4) Intact lumen/dilated lumen was (0 74±0 03) vs (0 35±0 07), P<0 01; (5)Effect of serum taken from group 1 on cultured smooth muscle cells showed by 3 H TdR method was (4110 14±1977 01) cpm, which was significantly less than that of serum taken from group 2 〔(7960 14±2802 59) cpm,P<0 05〕. Conclusion: BYHWD has significant preventive effect on the imitative post PTCA restenosis in diet induced atherosclerotic rabbits.
