摘要
用含有 0 .75mol/LNaCl的Zarrouk培养基洗去藻丝体表面的角质鞘 ,可获得大量有活性并能再生的单细胞 ,再生率为 2 8.6%。用 0 .4%的次氯酸钠溶液消毒藻丝体 5分钟 ,恢复培养后再置于含有终浓度均为 1 50 μg/μl的卡那霉素、庆大霉素和新霉素Zarrouk培养基中 35℃ 48h杀菌 ,最后用含 1 .5mol/LNaCl的Zarrouk培养基对藻丝体洗壁处理 1min 。
The present study suggested a lysozyme free method for preparation of single cells of axenic Spirulina platensis. When the filaments of Spirulina platensis washed by Zarrouk medium containing 0.75 mol /L NaCl, many single cells were obtained and the regeneration rate of single cells was 28.6%. A method is also explored for preparation of axenic strain of Spirulina platensis. Firstly, the filaments were treated with 0.4% NaClO for 5 minutes, secondly, put the filaments into the Zarrouk medium containing Kanamycin, Gentamicin and Neomycin, the final concentration of these three antibiotic is 150μg/μl, cultured the filaments under 35℃ for 48 hours, finally the filaments were rinsed with Zarrouk medium containing 1.5 mol/L NaCl for 1minute.
