摘要
目的 :研究缺血再灌注对脑组织水肿和ATP酶活性的影响。方法 :闭塞大鼠四条动脉造成急性全脑缺血 ,松开颈总动脉夹闭制成再灌注模型 ,以组织学方法观察缺血再灌注后脑组织水肿变化 ,同时测定脑组织中ATP酶活性和丙二醛 (MDA)的含量。结果 :①缺血 30min组与假手术组的组织学结构基本相同 ,仅出现软脑膜毛细血管轻度充血 ,神经细胞无明显肿胀 ,形态结构亦无损伤性改变 ,胶质细胞分布正常 ,而缺血再灌注组神经元和胶质细胞显著肿胀 ,部分神经细胞变性坏死。②与假手术组比较 ,缺血 30min组脑组织胞液和膜ATP酶活性均显著下降 ,再灌注 2 4h后酶活性进一步下降。③与假手术组比较 ,缺血 30min组脑组织胞液和膜MDA含量显著升高 ,再灌注 2 4h后MDA含量继续升高。结论 :缺血及再灌注造成的神经元损伤与能量代谢。
Objective:Study the influence on rat brain edema and ATPase activity with cerebral ischemia reperfusion.Method:Using the rat model with four artery occlusion and common carotid artery reperfusion,we observed the change of brain edema after ischemia and reperfusion;determined the ATPase activity and methylene dialdehyde(MDA)content at the same time.Result:①the histological structure of the group of ischemia 30 minutes is not different from the sham operate group;only capillary were slight hyperemia.There were no remarkable turgescence and change of morphological structure in nerve cell.The distribution of glial cell were normal.In ischemia reperfusion group nerve cell and glial cell were turgid;a part of nerve cell were degeneration,necrosis.②compared with the sham operate group,ATPase activity of brain cell sap and membrane decreased significantly in the group of ischemia 30 minutes and decreased continuously after reperfusion 24 hours.③compared with the sham operate group,MDA content increased remarkably in the group of ischemia 30 minutes and increased continously after reperfusion 24 hours.Conclusion:neuron injury induced by ischemia reperfusion were relative to energy metabolism,lipid peroxidation and deregulation of positive ion concentration between the inside and the outside of membrane.
