摘要
目的 探讨血管内皮生长因子 (VEGF)对牛眼视网膜色素上皮 (RPE)细胞DNA合成的影响 ,并观察榄香烯注射液的抗增殖作用。方法 采用3H -TdR掺入法 ,在体外培养的牛眼RPE细胞中加入不同浓度的VEGF以及VEGF和榄香烯 ,应用液闪仪测定3H TdR掺入的脉冲数 (CPM)。结果 不同浓度的VEGF均使CPM值明显增加 ,且与VEGF浓度呈剂量依赖关系 ,不同浓度的榄香烯均使RPE细胞的CPM值明显下降 ,且与榄香烯浓度呈剂量依赖关系。结论 VEGF有促进RPE细胞的DNA合成的作用 ,榄香烯可抑制生长因子诱导的RPE细胞的DNA合成。
OBJECTIVE To investigate effect on VEGF in the cultured bovine RPE cells as well as the anti-proliferative effect of elemene METHODS VEGF or VEGF and elemene with different concentration were put in the cultured bovine RPE cells by 3H-TdR incorporation method, then the CPM value of 3H-TdR incorporation were determined according to liquid scintillation counter RESULTS VEGF of different concentration made the value of CPM increase significantly in a dose dependent manner Elemene of different concentration made the value of CPM of PRE cells decrease significantly in a dose dependent manner CONCLUSIONS VEGF promoted DNA synthesis of RPE cells and elemene inhibited the DNA synthesis of RPE cells which was induced by VEGF
