-80℃冰箱非程控降温冻存脐血造血干细胞

摘要 目的:为脐血造血干细胞(CBHSC)的冻存建立简便、经济且有效的方法。方法:采用5%二甲基亚砜(DMSO)加6%羟乙基淀粉(HES)为冷冻保护剂,不用程控降温装置,直接置CBHSC于-80℃冰箱中冻存1~6个月。结果:冻存6个月后,单个核细胞(MNC)、粒-单系造血祖细胞(CFU-GM)和CD_(34)^+细胞的回收率及台盼兰拒染率分别为(91.6±1.8)%,(83.1±15.6)%,(88.2±1.8)%,(77.7±2.0)%。结论:这种简便、经济的方法能很好地保存CBHSC的造血潜能,因此,能为脐血移植冻存CBHSC。 Objective : To establish simple, inexpensive and effective methods for the cryopreservation of cord blood hematopoi-etic stem cells (CBHSC) . Methods: simplified methods were used to place CBHSC directly in a - 80℃ electrical freezer with 5% dimethylsulfoxide (DMSO) and 6% hydroxyethyl starch (HES) without rate-controlled freezing for 1 - 6 months. Results: For 6 months cryopreservation of cord blood mononuclear cells ( MNC), the mean recoveries of MNC and granulocyte macrophage colony forming units (CFU- GM) and CD34+ cells, and the mean trypan blue exclusion rate were,respectively, (91.6±1.8)%,(83.1± 15.6)%,(88.2±1.8)%,(77.7±2.0)%. Conclusion:This simple and inexpensive method can excellently maintain prolifera-tive capacity of CBHSC. Furthermore > they can be used to cryopreserve CBHSC for CBHSC transplantation.
