摘要
对辽宁、河南、湖北、湖南、福建和安徽各地引进的9个茯苓[Poria cocos(Fr)Wolf]菌株进行了12次筛选,结果证明,用PDA加20克/升松根煮液的培养基、在28℃下培养的编号5—(湖北)和编号6—(安徽)菌株,生长快、质地好。将转管3—4次、培养达20天的菌种,置于28℃以上、湿度85%以上,继续培养7—10天,形成子实体,进行有性繁殖,基本上解决了茯苓无性繁殖菌种易老化的问题。从各菌株的酯酶同工酶酶谱分析可以看出:有性繁殖中编号1菌株,具强、中、弱三条酶带、酶的活性强,在各菌株中最为优良。
The tuckahoe, Poria cocos (Fr. )Wolf, tends to degenerate after being cultured for 3-4 generations (asexual reproduction) on medium, a decrease in yield and quality. Long ago, many scientists had respected that screening fine strains and probe into a practical method could be resolved as mentiened above problems. Since 1985 screening to 9 strains introduced from Liaoning, Hubei, Hunan, Fujian and Anhui provinces has been made for 12 times. The results grow show that strain NO 5 (Hubei) and No 6 (Anhui) rapidly and the qualities are fine when cultured on PDA with 20g/1 boil liquid of pine root at 28℃, In order to overcome the degeneration caused by asexual reproduction, the old curtures incubated for 20 days and subcultured for 3-4 generations were re-incubated at above 28℃ and RH 85% for 7-10 days, the fruiting bodies formed on plate or in test tube, and then, the basidiospores were collected for sexual reproduction. Application of this method resolved (?)cally the degeneration caused by asexual reproduction. Besides the characteristics of strains, esterase isoenzymes were tested. The results indicate that strain No 1 has 3 enzyme bands (including strong, mid, weak) on enzyme pattern, and the enzyme activity is stronger than others, and proves to be the fine strain for the strain screening.
