水稻早恢402体细胞培养初步研究

对水稻品种早恢 40 2进行体细胞培养 ,结果表明 :(1)幼苗茎段比成熟种胚具有较高的愈伤组织诱导率 ,但前者绿苗分化率极低。 (2 )对幼苗进行 8℃ 2～ 3天的低温预处理 ,茎段愈伤组织诱导率虽有一定下降 ,但绿苗分化率却能大幅度提高。 (3)进行分化培养的愈伤组织进行 4～

Stem segments of the seedlings and mature embryos of the rice cultivar “Zao Hui 402” were used as explants in somatic tissue culture.Stem segments had higher callus induction rate than mature embryos,but their green shoot differentiation percentage was very low.Pretreatment of the seedlings at 8℃ for 2～3 days significantly raised the green shoot differentiation percentage from the cultured stem segment explants,though their callus induction percentage was decreased to some extent.Desiccation pretreatment of the calli for 4～6 days prior to shoot differentiation markedly improved green shoot differentiation percentage.
