摘要
目的 探讨孕酮 (PROG)减轻缺血 /再灌注 (I/ R)时脑水肿的机制。方法 采用大鼠局灶性脑 I/ R模型 ,分光光度计定量测定大脑中动脉阻塞 (MCAO) 2 h再灌注 2 2 h后脑皮层伊文思蓝 (EB)含量的变化及 PROG的影响。结果 MCAO侧 EB含量 I/ R组为 5 .89± 1.37μg/ g湿重 ,溶剂 (DMSO)组为 5 .0 3± 2 .70μg/ g湿重 ,PROG组为 2 .0 7± 0 .96μg/ g湿重 ;PROG组显著低于 I/ R组和 DMSO组 (P <0 .0 5 )。结论 PROG显著降低缺血 2 h再灌注 2 2 h时血 -脑脊液屏障 (BBB)的通透性 ,这可能是其减轻 I/
Objective To explore mechanism of progesterone alleviating brain oedema during ischemia/reperfusion(I/R).Methods The model of focal cerebral ischemia was established in rats by reversible inserting a nylon thread with a diameter of 0.2 mm into the anterior cerebral artery through the internal carotid artery.Content change of Evan’s blue(EB) in cortex subjected to 2 h middle cerebral artery occlusion(MCAO) followed by 22 h reperfusion and effect of progesterone on its was quantitatively determined by spectrophotometry.Results The EB content of cortex in MCAO side was 5.89±1.37μg/g wet wt in I/R group, 5.03±2.70μg/g wet wt in DMSO group, 2.07±0.96μg/g wet wt in PROG group. Compared with that in I/R and DMSO group,the EB content in PROG group was significantly reduced.Conclusion PROG could markedly lower permeability of blood brain barrier subjected to 2 h MCAO followed by 22 h reperfusion,and this may be a mechanism of PROG alleviating brain oedema during I/R.
