葡萄糖与果糖异构化的酶反应动力学 Ⅰ.实效动力学参数的测求

摘要 用国产异构化酶在固定床中研究了D-葡萄糖与D-果糖异构化反应动力学。利用米氏方程测定了60~75℃下正、逆异构化反应的米氏常数、最大反应速度与各机理反应的活化能、指前因子以及相对的速度常数等实效动力学参数与酶中间物的离解常数。利用计算标准摩尔反应焓与预测反应初期产物浓度对测求的实效动力学参数给以验证,计算值与实验值取得很好的吻合。 The kinetics of the isomerization of D-glucose and D-fructose was studied on the immobilized enzyme. The Michealis constants, maximum rates of the forward and reverse reaction and relative rate constants, activation energy, frequency factor of each reaction in the reaction mechanism were obtained by Kichealis-Menten equation over the temperature range of 60℃ to 75℃. The validity of these effective kinetics parameters may be confirmed by calculations of change of the enthalpy of reaction and concentrations of product at initial stage of the reaction. The values obtained agreed well with that calculated.
