局灶性脑缺血大鼠IL-8的变化

摘要 目的 :探讨 IL- 8在脑缺血损伤中的变化。方法 :采用改良 Zea Longa线栓法大鼠 MCAO模型 ,将大脑中动脉 ( MCA)闭塞不同的时间。应用双抗体夹心间接 ELISA法检测缺血组和对照组大鼠受伤脑及血清中 IL- 8浓度。结果 :闭塞 3h缺血侧半球湿重大于非缺血侧 ( P<0 .0 5)并维持至 48h;缺血组脑组织 IL- 8含量在 MCA闭塞 3h处于较低水平 ,随后逐渐升高 ,至 6h达高峰 ,随后又降低 ;血清中 IL- 8含量在缺血 3h达高峰 ,随后缓慢下降。空白组、假手术组脑组织内 IL- 8含量高于缺血相 ( P<0 .0 1 ) ,同一大脑非损伤侧高于损伤侧。结论 :IL- 8具有双重作用 ,既参与脑组织的正常代谢及生理功能 ,又参与了脑缺血损伤的病理过程。 Objective:To study the change of interleukin 8(IL 8) level in rats with cerebral ischemia injury.Methods:The middle cerebral artery(MCA) was occluded for different time with modified Zea Longas MCAO model.The level of IL 8 in injured brain tissue and blood serum was detected with double antibody sandwich indirect enzyme linked immunosorbent assay,both in ischemia group and in control group.Results:The weight of the injured bain hemisphere is heavier than that of uninjured hemisphere from 3 hours to 48 hours after cerebral ischemia ( P <0 05).The content of IL 8 in injured brain tissue was in lower level at 3 hours after ischemia,and then increased,peaked at 6 hours,and after this decreased.The concentration of IL 8 in serum peaked at 3 hours after ischemia,and then decreased slowly.The content of IL 8 in brain tissue of control group was higher than that in ischemia group ( P <0 01).Conclusion:IL 8 has double roles:it plays a role in metabolism and function in normal brain tissue and a role in pathological course of cerebral ischemia injury.
