摘要
目的研究P53抗癌基因突变与脑星形细胞瘤恶性程度的关系。方法采用非同位素PCR-SSCP分析法检测了31例星形细胞瘤患者P53抗癌基因的突变情况。结果(1)9例存在突变,突变区域分布于第5~8外显子中,以第6外显子突变率最高,第9外显子未测得突变病例。(2)2例存在多点突变,其中1例同时存在第5、6外显子突变,另1例同时存在第6、7外显子突变。(3)病理级别为Ⅰ、Ⅱ级组的突变率为0,Ⅲ级组的突变率为36.4%,Ⅳ级组的突变率为50%。Ⅲ、Ⅳ级组与Ⅰ、Ⅱ级组的突变率比较,均存在极显著差异(P<0.01)。
Objective To study the relationship between P^(53) tumor suppressor gene mutations and the malignant degree of brain astrocytomas.Methods Using nonisotope PCRSSCP molecular biological techniques,we detected the P^(53) tumor suppressor gene mutations in 31 specimens of astrocytomas.Results (1)Of all the 31 cases,P^(53) tumor suppressor gene mutations locating in exon 5~8 were detected,mutations were detected in 9 cases.Mutation rate in exon 6 was the highest,with no mutation being detected in exon 9.(2)Multiple point mutations were detected in 2 cases,one in exon 5~6 and the other in exon 6~7.(3)Mutation rate of pathological grade Ⅰ~Ⅱ group was O,that of grade Ⅲ group and grade Ⅳ group were 36.4% and 50%,respectively.The discrepancy of mutation rate between gradeⅠ~Ⅱ group and grade Ⅲ~Ⅳ group were significant (P<0.01).Conclusion The investigation demonstrated that P^(53) tumor suppressor gene mutation had a close relation to the malignant degree of astrocytomas.
