摘要 目的探讨维生素E对体外培养的人眼小梁细胞糖胺多糖(glycosaminoglycan,GAG)代谢的影响。方法第3~5代人眼小梁中加入100、200、400、800μg/ml的VitE,分离提纯后GAG经腊酸纤维薄膜电泳、闪烁测量测得人眼小梁细胞GAG的总量和各组份构成比。结果100、200、400μg/mlVitE能增加GAG的总量,同时增加透明质酸的构成比,降低含硫GAG的构成比。800μg/mlVitE降低GAG总量而不改变其构成比。结论VitE似有在发病学上防治POAG的可能性。 ObjectiveTo study the influence of Vit E on glycosaminoglycans (GAG) metabolism of cultured human trabecular meshwork cells.MethodsVit E in different concentrations of 100,200,400 and 800 μg/ml medium were added to the cells to the third to fifth generation.After extracting and purification,an aliquot of GAGs was used for cellulose electrophoresis and scintillation counting to determine the total amounts and their composition.Results100,200,400 μg/ml Vit E increased the total GAGs in the human trabecular meshwork cells.They also increased hyaluronic acid and decreased sulfatedGAGs.800 μg/ml Vit E descreased the total GAGs but keep the composition.ConclusionVit E can influence trabecular meshwork cells GAG metabolism.It is suggested that Vit E may be utilized to reverse the pathogenesis of POAG.
