摘要
目的研究K562细胞a、β、Gγ、Aγ、ζ、ε、δ、珠蛋白基因mRNA水平。方法采用RT-PCR在相同条件下同时扩增成 功7种珠蛋白mRNA,比较K562细胞内各珠蛋白mRNA水平。结果α基因簇中α-mRNA水平明显高于ζ-mRNA,β 基因簇mRNA表达水平依次为Gγ、Aγ、ε、δ,K562细胞不表达β-珠蛋白mRNA。结论转录后水平的调控可能在k562 细胞α基因簇的表达中起重要作用,β基因簇各珠蛋白mRNA水平和蛋白质水平基本相符。
Objective: To investigate theα、β、Gγ、Aγ、ζ、ε、δ、globin mRNA levels in K562 cells. Methods The technique of RT-PCR was applied successfully to amplify 7 globin mRNA in K562 cells, And comparative study of the relative amount of the 7 globin mRNA was performed. Result The expression level of α-mRNA in a globin gene cluster was higher than that of ζ-mRNA. The order of mRNA levels in β globin gene cluster was . β mRNA was not detected in K562 cells. Conclusion The expression level of mRNA in β globin gene cluster is consistent with that of β-like globin, the post transcriptional up- and down-regulation might be crutial in globin gene expression.
