摘要
用国产抗人角蛋白单克隆抗体,通过ABC免疫组化染色,观察T56细胞和HeLa细胞间期及有丝分裂期Keratin的组织结构和动态变化;并进一步观察了秋水仙素和细胞松弛素B对该两种细胞Keratin组织结构的影响。结果表明:有丝分裂时Keratin的组织结构及空间分布均发生显著变化;秋水仙素和细胞松弛素B共同作用时可引起Keratin纤维急剧重排形成一“星点状”纤维网络。
Avidin-Biotin Peroxidasecomplex technique, ABC staining, was em-loyed by using monoclonal anti-keratinantibody HK_2 in this study. The organ-ization and dynamics of keratins in bothinterphase and mitotic T56 and HeLa cellswere analysed. We also observed the ef-fects of microtubule (MT)and microfilament(MF)inhibitors, colchicine and cytochala-sin B, on the organization of keratin fila-ments in T56 and HeLa cells. The resultsshowed that a significant alteration in thestructural organization and distribution ofkeratin filaments occurred during mitosis,and an extensive rearrangemend of keratinnetworks of the two cell lines was in-duced in interphase after the MT and MFwere disrupted by combined treatmentwith the two drugs, colchicine and cyto-chalasin B;the keratin networks turnedinto a star-like lattice rapidly within1-2h.Neither colchicine nor cytochalasinB alone elicited significant organizationalchange in the keratin networks of the twocell lines.
