摘要
目的使肺癌细胞表达共刺激分子,探讨其作为疫苗的可能性。方法小鼠骨髓细胞在体外经GM-CSF和TNF-α诱导,使之成为成熟树突状细胞(BmDC)。将其与小鼠Lewis肺癌细胞系AL9901融合,并以S-P免疫细胞化学染色法检测融合细胞和BmDC上有关分子的表达。结果通过GM-CSF和TNF-α诱导获得BmDC,以诱导5d时收获为佳。获得的BmDC能高表达B7-1和CD40分子,与肺癌细胞融合后,获得的融合细胞仍能高表达B7-1和CD40分子。结论通过将小鼠肺癌细胞与树突状细胞融合,使肺癌细胞获得了BmDC表达的B7-1和CD40分子,从而提高了其免疫原性,为制备肺癌疫苗奠定了基础。
Aim To ameliorate expression of costimulatory molecules in lung cancer cells,so that the lung cancer cells could be used more effectively as vaccine. Methods The murine bone marrow(Bm) were induced with GM CSF and TNF α into dendritic cells(DC). There was a fusion cell strain obtained from murine Lewis lung cancer AL9901 cells and the BmDC. Molecule expression of the fusional cells and BmDC were detected by S P immunocytochemical staining. Results The mature murine BmDC can be induced with GM CSF and TNF α in vitro, and the BmDC grows best on 5d after induced. Five clone fusional cells have been obtained. The BmDC and the fusional cells can express strongly B7 1 and CD40 molecules, but LA9901 cells can′t do so. Conclusion The lung cancer cells can obtain BmDC molecule information after being fused with BmDC, then expression of B7 1 and CD40 in lung cancer cells can be ameliorated. Perhaps immunogenicity of fusional cell can be increased and used as vaccine. It is a good basis established for obtaining effective vaccine to control lung cancer by stimulating T cell immune.
