摘要
本文介绍一种用FOX法测定血浆中脂质过氧化的初级产物——脂质氢过氧化物(ROOH)的分光光度法。ROOH在稀酸溶液中可氧化Fe^(2+)变为Fe^(3+),后者与二甲酚橙高度结合产生一种蓝紫色的复合物,在分光光度计上于560 nm测定光密度,根据H_2O_2标准曲线可获得血浆中 ROOH值。该法灵敏度高,重复性好,所需设备简单,可望成为研究自由基和脂质过氧化反应的重要方法之一。
Ferrous Oxidation in Xylenol Orange (FOX) is a spectrophotometry to detect primary product of Lipid peroxidation in plasma - - Lipid Hydroperoxides(ROOH)In acid dilute solution,ROOH can oxidate Fe2+ into Fe3+ ,and Fe3+ can combine with Xylenol Orange to produce a kind of bluish - purple compound. On spectrophotometer the optical density is 560 nm. According to H2O2 standard curve,the value of ROOH in plasma can be detected. The sensitivity and repetivity of the method are good,and required equipment is simple,so it is expected to be a major method to study free radical and lipid peroxidation.
